SDM is a technique, which enables introducing desired targeted point mutations in the gene of interest. Of the many applications, the technique is of special importance to study the structure and function of protein. SDM can introduce a nucleotide substitution, deletion or addition. It is even possible to introduce deletion or insertion of a short stretch of DNA. The technique relies on the principle of providing a template sequence with the intended change in the primers. Using a pair of primers carrying the desired mutation to be introduced in the double stranded DNA, it is possible to alter the protein coding sequence as per the requirement.
